Biosynthesis of lactones from diols mediated by an artificial flavin

Abstract Background Lactones are important compounds in the field of medicine, material and chemical industry. One promising accesses to these flexible scaffolds is NAD(P) + -dependent alcohol dehydrogenases-catalyzed oxidative lactonization diols, which relies on construction an efficient regeneration system. Results In this study, a novel system combining horse liver dehydrogenase (HLADH) with synthetic bridged flavin cofactor was established for biosynthesis lactones. The reaction conditions were optimized variety lactones including chiral efficiently obtained from various diols. Compared previously reported -regeneration systems, showed better efficiency product yield. A two-phase further applied solve problem inhibition, 80% yield at condition 300 mM substrate. Conclusions This study provides method synthesis diols under mild conditions. We believe will be alternative promote other valuable compounds. Graphic abstract
